Why is 16S rRNA sequencing important?
The accurate species identification by 16S rRNA gene sequencing is also an important means to better define inter- and intraspecies phenotypic and genotypic variations, through studies on atypical bacterial isolates. Unusual strains of various Gram-positive and Gram-negative bacteria have been recognised by 16S rRNA gene sequencing (To et al., 2011; Woo, Cheung, et al., 2001; Woo, Leung, Leung, et al., 2001; Woo, Leung, et al., 2000; Woo, Teng, et al., 2003). For example, using 16S rRNA gene sequencing, a case of endocarditis and pericarditis caused by catalase-negative S. aureus was defined, which has led to the finding of a novel nonsense mutation in the katA gene encoding the catalase protein (To et al., 2011 ). 16S rRNA gene sequencing has also been applied to define rare infections such as a case of Haemophilus quentini bacteraemia leading to the identification of genetic mechanisms for reduced levofloxacin susceptibility and tetracycline resistance (Mak et al., 2005 ).
How does PCR work?
The PCR allows the millionfold amplification of a DNA fragment which is framed from two primers. In the first step, the template DNA that serves as a master copy is denatured by application of heat, typically at 95°C , which leads to the separation of the complementary DNA bases and the formation of two single strands.
