What is the purpose of ECL in Western blot?

ECL Western Blot Enhanced chemiluminescence (ECL) is a method which provides highly precise detection of proteins from Western blots. In many luminescenct assays, the light emitted is of low intensity and short duration. We use an enhanced version of the chemiluminescence reaction.

How does ECL work?

ECL uses an immunoassay format, with an antibody and labeled paramagnetic bead that captures the neurotoxin. A second detection antibody is used that is labeled with a chelate. When toxin is present, the detection and capture antibodies form an immunocomplex.

Excessive, diffuse signals

Excessive amounts of protein or high concentrations of antibodies, but also excessive exposure times, can lead to saturated signals. These signals are no longer proportional to protein concentration and may therefore not be used for quantitation of molecular weight.

Negative bands on the film

Occasionally, when imaging a membrane with chemiluminescence, white (negative) bands can appear alongside the normal black bands (Fig 3). These are the result of substrate depletion and it becomes difficult to quantitate the band in question.

Spotted (speckled) background

Spotted backgrounds (Fig 4) are a problem that can be caused by problems with the blocking agent, antibody, or contaminated equipment.

High background

Once again, there are several reasons why the background can be higher than you want it to be (Fig 5). This might result from insufficient blocking time or low concentration of blocking agent. Simply increase the blocking time and/or concentration of blocking agent.

What are the disadvantages of ECL?

One disadvantage of ECL methods is the frequent fouling of the electrodes. This effect can sometimes be prevented by regular electrochemical cleaning of the electrodes. In the case of an inexpensive electrode material such as aluminum, or even silicon manufactured in large quantities, or printed suitably treated printed conductors the electrode or even the whole bioaffinity assay cartridge containing the electrode chip is disposable.

How is hydrogen peroxide detected?

In indirect ECL methods, hydrogen peroxide is generated electrolytically at a negatively biased glassy carbon or gold electrode and detected through the chemiluminescence of, e.g., luminol. Hydrogen peroxide is transported by a liquid flow toward the chemiluminescent reagent and during this time is partially decomposed by the sample molecules, which act as a catalyst. The sample could be, for instance, heme components, which effectively catalyze the decomposition of hydrogen peroxide.

ECL can be more sensitive than fluorescence, but this is due to the nature of the detection method, not to the primary antibody.

What is ECL based on?

Basically, ECL is based on antibodies that are conjugated or labeled with horseradish peroxidase (HRP). The application of a chemiluminescent substrate such as luminol or acridan and a strong oxidizing agent such as hydrogen peroxide to the blot produces excited intermediates which then release a strong blue emission at 450 nm wavelength upon their decay to a lower energy level (ground state). Since light emission only occurs during the enzyme-substrate reaction, signal output ceases when the substrate in proximity to the enzyme is exhausted.
