They concluded that this may limit the use of SPME for quantitation in complex matrices. Inconsistent results with standardization methods also appear in SPME studies. For instance, Vaz Freire et al. 178 concluded that internal standards gave more reliable data than the method of standard additions, while Vianna and Ebeler 184 found the internal standard method to be unreliable and recommended the use of external standards. Ortega et al. 131 employed four internal standards, one for each of the major classes of trace components studied in their research in order to avoid the possibility of the internal standard behaving in a significantly different manner than the target components during extraction and separation. A validation of an HS–SPME for wine aroma components 81 confirmed that linearization of the response factor can be achieved with a suitable internal standard with a triphase DVB/CAR/PDMS fiber. SPME analysis of wines is complicated by the presence of 5–18% ethanol, which results in a rapid saturation of the fibers. This problem can be overcome by diluting the samples 5–10-fold in order to reduce the ethanol concentration below 2%. In this way, the least concentrated compound could, of course, fall below the detection threshold. Salt addition is recommended when least volatile and polar compounds must be included in the profile. A non-specific triphase DVB/CAR/PDMS fiber is generally suggested when the broader possible range of compounds is desired. Also, given the huge differences in the concentrations of different volatiles, the use of a suitable internal standard mix is recommended, as previously discussed. The most suitable extraction conditions generally depend on which class of compounds should be evidenced in the profile, since high temperatures and exposure times generally favor the least volatile and polar compounds, while the opposite applies for the most volatile and hydrophobic ones.

To solve the first problem a concentration using a stream of inert gas is used, but the more volatile compounds may be lost. The second problem is unsolved, unless one avoids the use of a solvent. This can be made using a thermal desorption unit, providing that the compounds are reversibly sorbed in the sorbent, and that the sorbent and the compounds are thermally stable.80
